抗SARS冠状病毒表面抗原抗体的制备及其初步应用

摘要: 研究探讨了制备和鉴定 SARS 冠状病毒表面抗原的快速免疫检测方法。采用基因工程表达SARS冠状病毒表面抗原免疫BALB/c小鼠,通过杂交瘤技术,获得了 4 株稳定分泌高特异性抗 SARS冠状病毒表面抗原的杂交瘤细胞系,制取了含高效价单克隆抗体的腹水;采用SARS冠状病毒表面抗原免疫家兔、绵羊、山羊,制取了抗SARS冠状病毒表面抗原的多克隆抗体。研究结果表明:CS4 C11单抗作为示踪抗体、绵羊抗SARS冠状病毒表面抗原抗体作为固相抗体为最佳配对;CS4 C11 单抗作为示踪抗体、CS6B12单抗作为固相抗体次之。获得的抗 SARS冠状病毒表面抗原的抗体为 SARS的早期诊断研究奠定了基础。Abstract: Four hybridoma cell lines secreting monoclonal antibody against SARS-associated coronavirus spike protein were obtained and at same time the polyclonal antibodies were also got by immunizing sheep, goats and rabbits with SARS-associated coronavirus spike protein respectively. Immunoradiometric assay(IRMA) and enzyme-linked immunosorbent assay(ELISA) were established using above Abs for detecting SARS-associated coronavirus, and best assay results were observed when McAb CS_4C_(11) was used as labeled Ab and PcAb(sheep) as capture Ab.
